If no one knows now, I sent an email to
golly-test@lists.sourceforge.net and attached a copy of my GollyPrefs file like dvgrn had suggested to me to help clear up any confusion about how the bug is reproduced. If any other information is needed to clear up any more problems, just suggest what additional information I'll need to provide.

Further generation count data: the file at Patterns/Life/Puffers/line-puffer-unstable.rle states

and states later on that

The exact generation is 9,746,067 for the defect; the last components of the puffer's "head" are destroyed at generation 9,746,134.
